Detection of cellulolytic fungi by using Congo red as an indicator: a comparative study with the dinitrosalicyclic acid reagent method
نویسنده
چکیده
Cellulolytic fungi can easily be screened within 2 d for the production of cellulolytic enzymes by staining with Congo red, or by measuring the amount of reducing sugar (glucose) produced with the dinitrosalicyclic acid reagent method. Endoglucanase activity is visible in carboxymethyl cellulose agar plates after staining with Congo red, and fixing with HCI or NaOH. This method is essentially based on the interaction of Congo red with intact P-(l-4)-~-glucans in carboxymethyl cellulose. Endoglucanase and exoglucanase activities of cellulases are quantitatively measured with the dinitrosalicyclic acid reagent method. The enzymatic activity detected with Congo red is compared with that obtained by the dinitrosalicyclic acid reagent method.

Isolation and Screening of Cellulolytic Fungi
A large number of microorganisms are capable of degrading cellulose, only a few of these microorganisms produce significant quantities of enzymes capable of completely hydrolyzing cellulose. Fungi are the main cellulase producing microorganisms. In this purposed study, four fungal species belonging to two genera i.e. Trichoderma and Aspergillus were isolated from different sources, screened and...
متن کاملDevelopment of Detection Methods for Cellulolytic Activity of Auricularia auricula-judae
To obtain basic information on the detection of cellulolytic activity in Auricularia auricula-judae, the influences of dye reagent, pH, and temperature were assessed. Chromogenic dye (congo red, phenol red, remazol brilliant blue, and trypan blue) was individually incorporated into a medium containing either carboxymethyl-cellulose, Avicel, or D-cellobiose as a polysaccharide carbon substrate. ...

متن کاملUse of Fenton Reagent as Advanced Oxidative Process for Removal of Basic and Acid Red Dyes from Aqueous Solutions
In this study, the Fenton process was used to investigate the color removal efficiency of Acid Red 52 and Basic Red 18 dye solutions by direct chemical oxidation. Color and chemical oxygen demand (COD) was successfully removed more than 99% and up to 92% respectively. This article would provide an easy assessment of the viability of using Fenton reagent in acidic conditions, so it is a prom...
متن کاملCatalytic Kinetic Determination of Silver through its Catalytic Effect on Congo Red-Peroxodisulphate Reaction
A kinetic-spectrophotometric procedure is reported for determination of trace amounts of Ag(I). This method is based on catalytic effect of silver ion on the oxidation of congo red by potassium peroxodisulphate at pH=3.4 and 30 oC in the presence of 1,10-phenanthroline as an activator. The reaction was followed spectrophotometrically by measuring the rate of change in absorbance ...
